Short Description: Bigfoot is a high-speed, high-throughput spectral cell sorter that acquires and sorts cells on spectrally unmixed or conventionally compensated data. It is used for analysis of complex multicolour samples, isolation of target cell populations, and single-cell deposition into plates.

Equipment Information:

Features:

  • High-speed jet-in-air cell sorter that is gentle on fragile cells and preserves viability.
  • Acquisition and sorting on spectrally unmixed or conventionally compensated data, from panel development to high-speed sorting.
  • Up to 6-way sorting into tubes and sorting into 96-, 384- and 1536-well plates.
  • Automated stream calibration and drop delay simplify sort setup; bubble detection stops the sort to protect precious samples.
  • Integrated Class II biocontainment cabinet and a separate aerosol management system protect the operator and the sample.
  • Applications include immunophenotyping, isolation of rare cell populations, single-cell sorting for genomics, cell line development and cloning.

Operating Principle:

  • Cells are hydrodynamically focused in a fluid stream and pass the laser beams one by one; light scatter and the full fluorescence spectrum are recorded and unmixed into the signals of individual fluorophores. The stream is broken into droplets, and droplets containing target cells are charged and deflected in an electrostatic field into tubes or plate wells.

Technical Specification:

  • Lasers (this configuration):
  • Detection channels (this configuration):
  • Platform capability: up to 9 lasers (349, 405, 445, 488, 532, 561, 594, 640, 785 nm) and up to 60 detectors, depending on configuration.
  • Analysis rate: >100,000 events/s; sort rate: >70,000 events/s.
  • Tube sorting: up to 6-way; holders for 1.5 mL, 5 mL, 15 mL and 50 mL tubes.
  • Plate sorting: 4-way into 96-well plates (in less than 8 s), 8-way into 384-well plates (in less than 11 s), straight-down into 1536-well plates.
  • Sample input: 1.5 mL, 5 mL and 15 mL tubes.
  • Interchangeable ceramic nozzle tips: 70, 100, 120 and 150 µm, with adjustable pressure.
