Short Description of the Internship Work: Short Description of the Internship Work: Lipid-based nanoparticles (LNPs) have many advantages compared to viral CRISPR/Cas9 delivery systems: ease of production, self-assembly, biocompatibility, high capacity, and many physicochemical properties that can be controlled to change their biological characteristics. Although the efficient delivery of CRISPR/Cas9 into living cells is still a major challenge.
The project aims to elucidate factors such as lipid composition, lipid charge, or curvature that may be involved in the molecular, biochemical, and biophysical events responsible for efficient cell entry and cargo delivery of gene editing tools or peptide-containing LNPs.
The project will create and form lipid nanoparticles of various compositions and sizes, which will be characterized by the DLS and HPLC methods, and their stability will be evaluated by the calcein release method or by the release of fluorescently labeled BSA.
