Preliminary Topic: Optimization of gene editing on human naive stem cells - cells with higher pluripotency

Short Description of the Internship Work: Naïve pluripotent stem cells (nPSCs) approximate the pre‑implantation epiblast, whereas “primed” PSCs resemble the post‑implantation epiblast. A cytokine‑independent “ground state” can be stabilized by dual inhibition of MEK/ERK and GSK3 (“2i”), establishing the canonical biochemical reference for naïve pluripotency. In humans, short‑term expression of NANOG/KLF2 coupled with small‑molecule conditions can reset primed PSCs to a naïve‑like state with transcriptomic and epigenetic features closer enhanced pluripotency.

In this project you will implement induced naïve pluripotent stem cells (inPSCs) cultures and optimize gene editing on them.
